Tackling antibiotic resistance: new antimicrobials through nanotechnology and protein engineering
The Margarita Salas Biological Research Center of the Higher Council for Scientific Research (CIB-CSIC, Madrid) seeks candidates with motivation and initiative to access one of the Grants for pre-doctoral contracts for the training of doctors 2022 from the State Research Agency (AEI). Candidates will acquire a comprehensive and complete training plan, with a set of multidisciplinary competence that include, from the understanding of the structure and folding of proteins, to nanotechnology and microbiology, with the final objective of applying the results in the biomedical field.

Title: Solenoid beta-beta domains as tools for molecular recognition of polymers: use as antimicrobials of respiratory pathogens and in dental health.

Resume: The structure beta-beta solenoide (BBS) it is a type of folding only studied in depth in pathogen surface proteins Streptococcus pneumoniae (pneumococcus). However, this same folding exists in several proteins involved in the recognition of various polysaccharides. The aim of this thesis is, firstly, to use native BBS modules (isolated or immobilized on nanoparticles) to control biofilms of dental plaque microorganisms by competition with the parental proteins from which they come, and secondly, to generate a library of mutants from which select new polypeptides that specifically recognize the surface of pathogens such as Streptococcus mutans O Pseudomonas aeruginosa. Estos dominios servirían para la creación "a la carta" de enzimas líticas híbridas de la pared bacteriana (enzymes) and that would make it possible to combat more effectively the growing phenomenon of antibiotic resistance. In all cases, the structural analysis of BBS-carbohydrate complexes by low-resolution (circular dichroism, fluorescence) and high-resolution (NMR) techniques will allow the molecular characterization of the type of interaction between the protein and its ligand and help in its application as antimicrobials. .

Title: Design of new antimicrobials by synthetic biology and nanobiotechnology: the bacterial envelope as a target.

Resume: Determinadas horquillas-β ("choline-binding repeats" o CBRs) pertenecientes al módulo de unión a colina C-LytA del patógeno respiratorio Streptococcus pneumoniae (pneumococcus)are capable of suffering unusual transition βa in the presence of micelles. Furthermore, these CBRs show antimicrobial activity against pneumococcus by destabilizing its membrane. The topic of the thesis consists of the biophysical study of the interaction of CBRs with minimal membrane systems (nanodiscs, microdroplets, microspheres) and in the investigation of their possible accumulation in vivo in intracellular microenvironments with phase separation, with a triple objective: i) Investigate the possible biological relevance of the aforementioned β transitiona; ii) utilizar mutantes de C-LytA que muestren una interacción mejorada con lípidos como "tags" para la functionalization of lipid nanoparticles with recombinant proteins, and iii) the use of C-LytA-derived peptides and polypeptides as antimicrobial agents against various pathogens. In parallel, the use of multivalent nanoparticles for the modulation / inhibition of processes of the division of Streptococcus pneumoniae mediated by FtsZ (the control of FtsZ assembly, the formation of biomolecular condensates, or the interaction with other elements of the division machinery).
